Isolation and Screening of Aspergillus niger and Bacillus coagulans as Potential Candidates for Amylase and Glucose Isomerase Production

There is an increasing demand for high fructose corn syrup as alternative to glucose especially use by people with diabetes due its low glycemic index. The cost of this product coupled has attracted the attention many researchers search and sustainable production route. Therefore, focus work was isolating screening Aspergillus niger Bacillus coagulans possible producers amylase isomerase respectively, which are needed produce syrup. Nine (9) samples loamy soil were taken from three (3) separate locations (BG = Botanical Garden, RD Refuse Dumpsite, FB Flower Bed) isolate bacterium fungus. Based on lacto-phenol cotton blue stain preparation under a microscope, colonies that thought be observed further identified. Cultural, microscopic, biochemical characteristics used confirm identification coagulans. probable isolates then screened respectively. Out total nine analysed, 5 (55.6 %) positive niger. A higher isolation rate niger, 2 (66.7 recorded in botanical garden (BG) flower bed (FB). lowest rate, 1 (33.3 obtained refuse dump. all their potential amylase, showed highest zone starch hydrolysis (28 mm), least (13 mm). On other hand, out 6 occurrence coagulans, 3 (100 (BG1), while occurrence, (FB2). revealed six (6) produced isomerase, dump (RD2) producing concentration (4.7014 g/L).
